Mitochondrial Channels and Their Role in Cardioprotection

نویسندگان

چکیده

Mitochondria play a pivotal role in cardioprotection. The major cardioprotective mechanism is ischemic preconditioning (IpreC), through which short periods of ischemia protect subsequent prolonged acute episode. channels, particularly the potassium channels (mitoK) such as ATP-dependent and calcium-activated have been suggested trigger or end effectors IpreC. Activators mitoK are promising therapeutic agents for treatment myocardial injury due to episodes. In this chapter, we summarizing our current knowledge on physiology function different mitochondrial with focus their Furthermore, currently under development therapy by targeting heart failure also discussed.

Role of the mitochondrial ATP-sensitive K+ channels in cardioprotection.

The mitochondrial ATP-sensitive K(+) (mitoK(ATP)) channel was discovered more than a decade ago. Since then, several pharmacological studies have identified agents that target this channel some of which selectively target mitoK(ATP). These and other studies have also suggested that mitoK(ATP) plays a key role in the process of ischemic preconditioning (IPC) and prevention of apoptosis. The mech...

Role of the Mitochondrial Permeability Transition in Myocardial Disease Evidence for Mitochondrial K Channels and Their Role in Cardioprotection Primary and Secondary Signaling Pathways in Cardioprotection Mitochondrial Death Pathways

Mitochondria play a key role in determining cell fate during exposure to stress. Their role during ischemia/reperfusion is particularly critical because of the conditions that promote both apoptosis by the mitochondrial pathway and necrosis by irreversible damage to mitochondria in association with mitochondrial permeability transition (MPT). MPT is caused by the opening of permeability transit...

Role of the Mitochondrial Permeability Transition in Myocardial Disease Primary and Secondary Signaling Pathways in Early Preconditioning That Converge on the Mitochondria to Produce Cardioprotection Evidence for Mitochondrial K Channels and Their Role in Cardioprotection Mitochondrial Death Pathways

Twenty years after the discovery of sarcolemmal ATP-sensitive K channels and 12 years after the discovery of mitochondrial KATP (mitoKATP) channels, progress has been remarkable, but many questions remain. In the case of the former, detailed structural information is available, and it is well accepted that the channel couples bioenergetics to cellular electrical excitability; however, in the he...

Opioid-induced second window of cardioprotection: potential role of mitochondrial KATP channels.

Opioids have been previously shown to confer short-term cardioprotection against a prolonged ischemic insult. Therefore, the present study was designed to determine whether opioids can induce a delayed or "second window" of cardioprotection and to assess the potential involvement of the mitochondrial KATP channel. All rats were subjected to 30 minutes of ischemia and 2 hours of reperfusion (I/R...

Mitochondrial ATP-dependent potassium channels: novel effectors of cardioprotection?

BACKGROUND Brief interruptions of coronary blood flow paradoxically protect the heart from subsequent prolonged ischemia. The basis of such endogenous cardioprotection, known as "ischemic preconditioning," remains uncertain. Pharmacological evidence has implicated ATP-dependent potassium (KATP) channels in the mechanism of preconditioning; however, the effects of sarcolemmal KATP channels on ex...
